De VriesPituitary pars intermedia dysfunction (PPID) is characterized by a progressing loss of dopaminergic inhibition of the pituitary pars intermedia and commonly causes poor physical appearance, hirsutism, and muscle loss in the geriatric horse. We hypothesized that horses with PPID have reduced tolerance to glucose and sensitivity to insulin, reduced mRNA abundance of SLC2A4, INSR, and SLC38A2, and reduced protein expression of GLUT4 and INSR in muscle tissue, reduced intestinal morphology, and reduced D-glucose and L- Gln transport capacity across the jejunal mucosa, and mRNA abundance of SLC5A1 and SLC38A2 compared to horses with no PPID. Horses were maintained on hay and senior feed for a period of 5 d while being monitored for medical stability. Presence and grade of PPID was determined based on endocrine testing and post mortem histopathological evaluation of the pituitary gland. Intestinal and gluteal muscle samples were harvested and processed. The results of this study show that muscle wasting and poor physical appearance associated with the pathology of the PPID condition is not due to a reduced transporter abundance at the intestinal and systemic level, but may be due in part to a reduced intestinal morphology. Geriatric horses and those with PPID in particular should be carefully managed with feeding diets containing low concentration of soluble carbohydrates in an effort to decrease the risk of laminitis.

Show moreIntegration of automatic milking systems (AMS) into Midwestern pasture-based systems has been suggested as an alternative low cost and labor efficient dairy system, but proper integration of this technology into a pasture-based dairy system is not well understood. The objective of this thesis was to determine management guidelines for a pasture-based AMS system by examining the combined effect of stocking rate and feeding strategies on cow performance, AMS use, feed degradation, dairy cattle genotype, and pasture utilization. An investigation lasting 8 weeks was conducted using a completely randomized design with a 2 x 2 factorial of two stocking rate systems ( high stocking rate (HSR 2.89 cows/ha); low stocking rate (LSR 1.92 cows/ha)) and two genotypes of Holstein cattle, United States Holstein (USH) and New Zealand Holstein Friesian (NZF). The HSR treatment received a partial mixed ration (PMR; 40% ground corn and 60% legume grass haylage) in variable amounts (average: 4.86 kg /cow; ± 0.14; mean and SEM) to supplement deficits in pasture growth rate and availability. Results indicated no differences in milk production or AMS use between treatments, although differences were found between genotypes. An in situ experiment showed no differences in feed degradation, suggesting that the rumen environment was not affected by PMR supplementation. No differences in pasture utilization or pasture growth were detected between the HSR and LSR treatments. The results of this thesis suggest that dynamic supplementation in the HSR treatment did not affect pasture utilization, cow performance, AMS use, or the rumen environment.

Though nasal epithelial remodelling has been well-documented in laboratory animals repeatedly exposed to ozone, associated inflammatory responses have not been fully characterized. In this study, we investigated if the onset of ozone-induced nasal epithelial remodelling is related to temporal changes in granulocytic influx and cytokine gene expression. Mice exposed to 24-weekdays of ozone developed marked eosinophilic rhinitis with epithelial hyperplasia, mucous cell metaplasia and hyalinosis. Repeated subacute ozone exposures in mice induced an eosinophilic rhinitis with epithelial remodelling that resembled the pathology of human allergic/non-allergic rhinitis. Ozone-induced eosinophilic rhinitis was associated with an initial T helper cell 1 (Th1), followed by Th2-inflammatory response. Based on these findings, a study was conducted to investigate the role of lymphocytes (hypothesized cellular sources of Th1- and Th2-cytokines) in the development of eosinophilic rhinitis and associated nasal epithelial remodelling. In Rag2 x common gamma chain (γc) - deficient [RAG2(-/-) x γc(-/-)] mice, which lack T- and B- lymphocytes as well as NK cells, ozone nasal epithelial remodelling and eosinophilic rhinitis did not develop, hence supporting the hypothesis that lymphocytes are a crucial component to ozone induced eosinophilic rhinitis and associated epithelial changes in mice. These results suggest that chronic exposure to air pollutants, like ozone, may contribute to rising prevalence of allergic and non-allergic rhinitis.

"Bacterial wilt on Lolium perenne and Poa annua caused by Xanthomonas translucens is a major concern on golf courses in the United States. Results revealed that the shortest generation time of each bacterial isolate was measured at 350 C, bacterial isolates were more pathogenic to the host from which they were isolated and varieties in virulence did exist in both L. perenne bacterial isolates and P. annua bacterial isolates. A positive correlation between temperature and disease severity was discovered on both L. perenne and P. annua. Results also revealed that L. perenne bacterial isolate, r3, is different from other bacterial isolates at many aspects. The 16S rDNA identification using a 1276-bp 16S rDNA sequence and the multilocus sequence analysis (MLSA) using a 1803-bp concatenated sequence of four housekeeping genes (gyrB, fusA, gapA and lepA) demonstrated that the L. perenne bacterial isolates, r0 and r5, were X. translucens pv. graminis and the P. annua bacterial isolates, p0, p6 and p8, were X. translucens pv. poae and suggested that the L. perenne bacterial isolate, r3, may be a new Xanthomonas species."--Page ii.
